低温条件下でタンパク質発現が可能なため、中温条件下で「宿主細胞にとって致死性があるタンパク質」、「宿主細胞の増殖を阻害するタンパク質」、「封入体を形成し凝集するタンパク質」などの組換えタンパク質を 発現・精製できることが期待できます。
Rhodococcus erythropolis L88 Competent Cells と pTip, pNit, pCpi Vector シリーズの組合せにて実現可能です。
Rhodococcus erythropolis L88 Competent Cells
製品仕様
Rhodococcus erythropolis L88 株はRhodococcus erythropolis JCM3201 株に対してUV 照射により点突然変異を導入した株であり、高いリゾチーム感受性を示します。通常、野生株は1 mg/mL 以上のリゾチームに耐性を有しますが、R. erythropolis L88 株では12.5 μg/mL のリゾチームにて溶菌を起こすことが可能です。そのため、細胞破砕が容易になりタンパク質の抽出および精製が野生株と比べて容易となります。
また、本株は4℃から30℃で生育を示し、4℃という低温条件下で外来遺伝子のコードするタンパク質を発現産生させることが可能です。

製品仕様
発現を制御するチオストレプトン誘導型プロモーター（pTip）、中程度の発現（pNit）、高発現（pCpi）の構成型プロモーターと、異なる複製開始起点(θ-type、rolling circle-type) をもつベクターとの組み合わせで、発現レベルの制御から共発現系まで多様な実験系をカバーすることが可能です。
導入タンパク質の発現実験を行う際には、Rhodococcus erythropolis L88 Competent Cells（ 製品コード：RE-L88）を宿主としてご利用いただけます。
R. erythropolis L88 株の形質転換効率は1×104 colonies/μg 以上であり、高い形質転換効率を示します。本製品はシャトルベクターであり、クローニングやコンストラクトの際には大腸菌を宿主として使用することができます。大腸菌を宿主とした場合、高コピーベクター（コピー数：300-500）として機能します。
両末端（あるいはそのいずれか一方）にヒスチジンタグが付加できるベクター構造となっているため、汎用カラムによる簡易精製にて、目的タンパク質を容易に精製することができます。
