Product Description

This is the custom polyclonal antibody production service provided by Hokudo Co., Ltd.

Antiserum production service is available not only for rabbit and mouse but also for large mammals such as goat and sheep.

Please prepare for an antigen sample and send it to us, and the works from immunization to serum collection will be performed.

The following works are included in the basic charge: immunization, pre-immunization blood sampling, test blood sampling, whole blood sampling, antibody titer measurement by ELISA and antiserum delivery.

Rabbit Polyclonal Antibody Production

Time Schedule Works
Pre-immunization Upon the consulting with the customer, the schedule will be provided.
Rabbit is arranged according to the schedule of receiving the antigen, and habituated for one week afterwards.
The concentration of the antigen should be 0.5mg/mL or more for protein antigen and 2mg/mL or more for synthetic peptide.
Freund’s adjuvant will be also used.
Week0 Pre-immunization serum (approx. 1.5mL) is harvested, then the first immunization is performed.
Week2 The second immunization is performed.
Week3 Test blood sampling and ELISA measurement are performed. The pre-immunization serum and antiserum (approx. 1.5mL) will be delivered.
Week4 Whole blood sampling is performed.

Amount of antigen administration per each work:
1st immunization　Protein: 0.2mg/rabbit, Synthetic peptide: 1mg/rabbit
2nd immunization　Protein: 0.1mg/rabbit, Synthetic peptide:0.5mg/rabbit

Required antigen amount(including the amount for possibly required additional immunization)

– Synthetic peptide:
For immunization: 5mg or more per rabbit (including the amount for additional immunization)
For ELISA measurement: 0.1mg (as a free peptide)
For immunization, please prepare a carrier protein binding peptide.

– Protein:
For immunization and ELISA measurement: 1mg or more per rabbit
In case of using a fusion protein, 0.1mg of either free protein or differently tagged fusion protein would be required for ELISA measurement.

In case that the absorbance value of 6,400 folds diluted 3w serum is 1.000 Abs or less, additional immunization is available up to twice for free if required.
(The additional 4 weeks animal breeding, test blood sampling and ELISA measurement are inclusive.)
Any additional immunization afterwards will be charged separately.

【For synthetic peptide antigen】

Time Schedule Works
Pre-immunization Upon the consulting with the customer, the schedule will be provided.
An animal is habituated for 4 weeks before starting immunization.
Week0 Pre-immunization serum (approx. 1.5mL) is harvested, then the first immunization (FCA) is performed.
Week2 The second immunization (FIA) is performed.
Week4 The third immunization (FIA) is performed.
Week6 The fourth immunization (FIA) is performed.
Week7 The first test blood sampling and ELISA measurement are performed. The antiserum (approx. 1.5mL) and the data will be delivered.
Week8 The fifth immunization (FIA) is performed.
Week9 Whole blood sampling is performed.

Administration amount of antigen is 5mg each per animal.

The required amount of antigen is 25mg or more per animal for immunization and 0.1mg (free synthetic peptide) for ELISA measurement.

Additional immunization will be charged separately.

【For protein antigen】

Time Schedule Works
Pre-immunization Upon the consulting with the customer, the schedule will be provided.
An animal is habituated for 4 weeks before starting immunization.
Week0 Pre-immunization serum (approx. 1.5mL) is harvested, then the first immunization (FCA) is performed.
Week2 The second immunization (FIA) is performed.
Week4 The third immunization (FIA) is performed.
Week5 The first test blood sampling and ELISA measurement are performed. The antiserum (approx. 1.5mL) and the data will be delivered.
Week6 The fourth immunization (FIA) is performed.
Week7 Whole blood sampling is performed.

Administration amount of antigen is 2mg each per animal.

The required amount of antigen is 10mg or more per animal for immunization and ELISA measurement.
In case of using a fusion protein, 0.1mg of either free protein or different type of fusion protein would be required for ELISA measurement.

Additional immunization will be charged separately.
